چکیده

Introduction Articular cartilage has a poor intrinsic capacity for repair. Bone marrow (BM) derived mesenchymal stem cells (MSCs) are an attractive cell type for use in cartilage tissue engineering due to their ease of expansion. A central challenge in tissue engineering is preventing core degradation. This challenge will be amplified if MSCs are to be used in the repair of large bone defects via endochondral ossification. We have recently shown that with a combination of modified scaffold architecture and rotational culture a more homogenous tissue using chondrocytes (CCs) can be engineered. The objective of this study is to investigate if this approach can be extended to BM derived MSCs.

Composition-function relations of cartilaginous tissues engineered from chondrocytes and mesenchymal stem cells isolated from bone marrow and infrapatellar fat pad.

The objective of this study was to determine the functional properties of cartilaginous tissues generated by porcine MSCs isolated from different tissue sources, and to compare these properties to those derived from chondrocytes (CCs). MSCs were isolated from bone marrow (BM) and infrapatellar fat pad (FP), while CCs were harvested from the articular surface of the femoro-patellar joint. Cultur...

Study of Chondrogenic Effects of Chondrocytes Cocultured With Murine Bone Marrow-Derived Mesenchymal Stem Cells

Purpose: Co-culture systems of marrow derived mesenchymal stem cells (mMSCs) with mature chondrocytes have theoretically been considered as a putative way of MSCs chondrogenic differentiation. MSCs differentiated in this system could be used for transplantation purpose without of any need to their purification since the cells with which MSCs are co cultured are native cartilage cells. Despite o...

Potential Use of Amniotic Membrane Derived Scaffold for Cerebrospinal Fluid Applications

Scaffolds derived from decellularized tissues provide a natural microenvironment for cell culture. Embryonic cerebrospinal fluid (e-CSF) contains factors which play vital roles in development of the nervous system. This research was aimed to survey the effect of Wistar rat e-CSF on neural differentiation of bone marrow derived mesenchymal stem cells (BM-MSCs) cultured on the human amniotic memb...

Altering the Architecture of Tissue Engineered Hypertrophic Cartilaginous Grafts Facilitates Vascularisation and Accelerates Mineralisation

Cartilaginous tissues engineered using mesenchymal stem cells (MSCs) can be leveraged to generate bone in vivo by executing an endochondral program, leading to increased interest in the use of such hypertrophic grafts for the regeneration of osseous defects. During normal skeletogenesis, canals within the developing hypertrophic cartilage play a key role in facilitating endochondral ossificatio...
